Various flavonoids have been shown to possess a variety of pharmacological activities and are found in abundance in plants, foods, and beverages. Previous studies have indicated that flavonoids may be useful in treating pulmonary disorders. We have shown previously that the flavonoids luteolin, apigenin, naringenin, quercetin, and genistein inhibit the growth of lung cancer cells and induce cell cycle arrest. In the present study, the mechanism of the anti-lung cancer activity of the flavonoids was investigated. Human lung cancer cells (A549) were treated with five flavonoids for 24h and the growth inhibition of the cells was measured. The cytotoxic activities of the flavonoids were in the order of apigenin > naringenin > quercetin > luteolin > genistein. Treatment of A549 cells with apigenin, naringenin, quercetin, and genistein induced G(2)-M arrest in a time-dependent manner. Moreover, we observed that the flavonoids blocked the growth of A549 cells by inhibiting the expression of aquaporin-1 (AQP1) mRNA and protein. The inhibition of AQP1 expression was also observed in cells treated with the flavonoids. These findings suggest that flavonoids inhibit AQP1 expression and this may, in part, mediate the cytotoxicity of flavonoids in A549 cells. FILED NOT FOR PUBLICATION MAR 08 2012
